У попередній статті ми описали шість білкових ролей, проти яких Rasayana перевіряє кандидатів докінгом — не називаючи самі білки, лише їхні функції (див. «Шість мішеней старіння»). Логічне питання від уважного читача: а чому взагалі вважати, що саме ці процеси пов'язані зі старінням, а не просто «звучать науково»? Ця стаття відповідає саме на це — з посиланнями на конкретні дослідження, а не на загальні враження.

Для кожної ролі є два принципово різні типи доказів, і варто їх не плутати. Перший — асоціативний: показник погіршується з віком у людей або пов'язаний з вищою смертністю (кореляція, не причинність). Другий — причинний: коли дослідники експериментально змінюють саме цей механізм у модельного організму (мушка, черв'як, миша) — і тривалість чи якість життя вимірно змінюється. Другий тип доказів набагато сильніший, і нижче за кожною роллю вказано, якого типу докази для неї є.

Шість ролей і докази за кожною

Фермент підтримки теломер

Добудовує захисні «ковпачки» на кінцях хромосом, які коротшають з кожним поділом клітини.

Асоціативний доказ: у людей від 60 років коротші лейкоцитарні теломери пов'язані з достовірно вищою смертністю в наступні роки (Cawthon et al., 2003). Причинний доказ: штучна реактивація ферменту в старих мишах з дефіцитом теломерази оборотно усуває ознаки тканинної дегенерації — атрофію органів, порушення нейрогенезу (Jaskelioff et al., 2011).

NAD⁺-залежна деацетилаза (сиртуїновий тип)

Регулює метаболічну «економність» клітини у відповідь на рівень енергії.

Асоціативний доказ: рівень NAD⁺ у тканинах людини вимірно падає з віком паралельно зі зростанням окислювального стресу (Massudi et al., 2012). Причинний доказ: активація цього шляху через підвищення NAD⁺ подовжує життя круглим черв'якам через мітохондріальну UPR-відповідь і FOXO-сигналінг (Mouchiroud et al., 2013).

Кіназа шляху чутливості до поживних речовин (mTOR-типу)

«Вирішує», чи клітина росте й ділиться, чи економить ресурси.

Причинний доказ (найсильніший у списку): рапаміцин, що пригнічує цей шлях, подовжив життя генетично різнорідним мишам навіть при старті лікування в похилому віці — знаковий результат програми NIA Interventions Testing Program (Harrison et al., 2009). Регулятор антиоксидантної відповіді (Nrf2-типу)

Керує «увімкненням» власної системи захисту клітини від окислювального стресу.

Асоціативний доказ: активність цього шляху знижується з віком, що послаблює клітинний захист від накопичуваного окислювального пошкодження (Zhang, Davies & Forman, 2015). Непрямий причинний доказ: у голих землекопів — гризунів з аномально великою тривалістю життя як для їхнього розміру — цей шлях працює помітно активніше, ніж у звичайних мишей (Lewis et al., 2015).

Ацетилтрансфераза, пов'язана з регуляцією автофагії

Одна з точок, де можна фармакологічно підкрутити «службу прибирання сміття» клітини.

Асоціативний доказ: автофагічна активність тканин знижується з віком практично в усіх вивчених модельних організмах (Rubinsztein, Mariño & Kroemer, 2011). Причинний доказ: миші зі штучно посиленою автофагією (надекспресія гена Atg5) живуть довше й демонструють кращі показники старіння тканин (Pyo et al., 2013).

Енергетичний сенсор (AMPK-типу)

Активується при нестачі енергії в клітині й запускає каскад «режиму економії».

Асоціативний доказ: активність цього сенсора послідовно знижується з віком у широкому діапазоні тканин (Salminen & Kaarniranta, 2012). Причинний доказ: подовження життя круглим черв'якам ефектом обмеження калорій вимагає саме цього сенсора — без нього ефект зникає (Mair et al., 2011).

Чому це все ще не доказ користі нашого кандидата

Ці дослідження доводять, що самі механізми — валідні мішені геронауки: змінюючи їх, справді можна вплинути на старіння модельних організмів. Але це не те саме, що довести, що конкретна речовина-кандидат Rasayana реально й достатньо сильно на них впливає в живій клітині — це вже питання докінгу (див. попередню статтю) і подальших лабораторних тестів, а не тільки літературного огляду.

Чому без назв генів. Як і в попередній статті, ролі описано узагальнено: маючи повний набір «речовина + білок + PDB-структура», нашу роботу можна було б відтворити без нас. Наведені тут дослідження стосуються ролі механізму в біології старіння загалом — вони опубліковані незалежно від Rasayana і не розкривають, які саме кандидати ми тестуємо.

In an earlier post we described six protein roles Rasayana tests candidates against via docking, without naming the proteins themselves — only their functions (see "Six Aging Targets"). A careful reader's next question: why believe these particular processes are actually linked to aging, rather than just "sounding scientific"? This post answers exactly that — with links to specific studies, not general impressions.

For each role there are two fundamentally different kinds of evidence, and they shouldn't be conflated. The first is associative: the marker worsens with age in humans, or correlates with higher mortality (correlation, not causation). The second is causal: researchers experimentally change that exact mechanism in a model organism (fly, worm, mouse) — and measurably change lifespan or healthspan. The second kind is far stronger evidence, and each role below states which type applies.

The six roles, and the evidence behind each

A telomere-maintenance enzyme

Rebuilds the protective "caps" on chromosome ends, which shorten with every cell division.

Associative evidence: in people over 60, shorter leukocyte telomeres are linked to reliably higher mortality in subsequent years (Cawthon et al., 2003). Causal evidence: artificially reactivating the enzyme in old telomerase-deficient mice reversibly resolves signs of tissue degeneration — organ atrophy, impaired neurogenesis (Jaskelioff et al., 2011).

An NAD⁺-dependent deacetylase (sirtuin-type)

Regulates the cell's metabolic "thriftiness" in response to energy status.

Associative evidence: NAD⁺ levels in human tissue measurably decline with age alongside rising oxidative stress (Massudi et al., 2012). Causal evidence: boosting this pathway via elevated NAD⁺ extends lifespan in roundworms through mitochondrial UPR activation and FOXO signaling (Mouchiroud et al., 2013).

A nutrient-sensing pathway kinase (mTOR-type)

"Decides" whether a cell grows and divides or conserves resources.

Causal evidence (the strongest on this list): rapamycin, which inhibits this pathway, extended lifespan in genetically heterogeneous mice even when treatment started late in life — a landmark result from the NIA's Interventions Testing Program (Harrison et al., 2009). An antioxidant-response regulator (Nrf2-type)

Controls the cell's own defense system against oxidative stress.

Associative evidence: activity of this pathway declines with age, weakening cellular defense against accumulating oxidative damage (Zhang, Davies & Forman, 2015). Indirect causal evidence: naked mole-rats — rodents with an anomalously long lifespan for their size — show markedly higher activity of this pathway than ordinary mice (Lewis et al., 2015).

An acetyltransferase linked to autophagy regulation

One point where the cell's "cleanup crew" can be pharmacologically nudged.

Associative evidence: autophagic activity in tissue declines with age across virtually every model organism studied (Rubinsztein, Mariño & Kroemer, 2011). Causal evidence: mice with artificially boosted autophagy (Atg5 overexpression) live longer and show better tissue-aging outcomes (Pyo et al., 2013).

An energy sensor (AMPK-type)

Activates when a cell is short on energy and triggers an "energy-saving mode" cascade.

Associative evidence: this sensor's activity consistently declines with age across a wide range of tissues (Salminen & Kaarniranta, 2012). Causal evidence: the lifespan extension calorie restriction produces in roundworms requires this exact sensor — without it, the effect disappears (Mair et al., 2011).

Why this still isn't proof our candidate helps

These studies prove the mechanisms themselves are valid geroscience targets: changing them really can affect aging in model organisms. That's not the same as proving a specific Rasayana candidate compound actually acts on them strongly enough in a living cell — that's a question for docking (see the previous post) and subsequent lab testing, not a literature review alone.

Why no gene names. As in the previous post, roles are described generically: with the full "compound + protein + PDB structure" dataset, our work could be reproduced without us. The studies cited here concern each mechanism's role in aging biology in general — published independently of Rasayana, and revealing nothing about which specific candidates we're testing.

Die nächste Frage eines aufmerksamen Lesers: Warum sollte man annehmen, dass genau diese Prozesse mit dem Altern verbunden sind, statt nur „wissenschaftlich zu klingen"? Genau das beantwortet dieser Beitrag — mit Verweisen auf konkrete Studien, nicht auf allgemeine Eindrücke.

Für jede Rolle gibt es zwei grundlegend verschiedene Arten von Belegen, die man nicht verwechseln sollte. Die erste ist assoziativ: Der Marker verschlechtert sich mit dem Alter beim Menschen oder korreliert mit höherer Sterblichkeit (Korrelation, keine Kausalität). Die zweite ist kausal: Forscher verändern genau diesen Mechanismus experimentell in einem Modellorganismus (Fliege, Wurm, Maus) — und Lebensspanne oder Gesundheitsspanne ändern sich messbar. Die zweite Art ist ein weitaus stärkerer Beleg; bei jeder Rolle unten steht, welcher Typ zutrifft.

Die sechs Rollen und ihre jeweiligen Belege

Ein Enzym zur Telomer-Erhaltung

Baut die schützenden „Kappen" an den Chromosomenenden wieder auf, die sich mit jeder Zellteilung verkürzen.

Assoziativer Beleg: Bei Menschen über 60 sind kürzere Leukozyten-Telomere mit zuverlässig höherer Sterblichkeit in den Folgejahren verbunden (Cawthon et al., 2003). Kausaler Beleg: Die künstliche Reaktivierung des Enzyms bei alten, telomerasedefizienten Mäusen behebt reversibel Anzeichen von Gewebedegeneration — Organatrophie, gestörte Neurogenese (Jaskelioff et al., 2011).

Eine NAD⁺-abhängige Deacetylase (Sirtuin-Typ)

Reguliert die metabolische „Sparsamkeit" der Zelle je nach Energiestatus.

Assoziativer Beleg: Der NAD⁺-Spiegel im menschlichen Gewebe sinkt mit dem Alter messbar, während oxidativer Stress zunimmt (Massudi et al., 2012). Kausaler Beleg: Die Aktivierung dieses Signalwegs durch erhöhtes NAD⁺ verlängert das Leben von Fadenwürmern über mitochondriale UPR-Aktivierung und FOXO-Signalgebung (Mouchiroud et al., 2013).

Eine Kinase des Nährstoff-Sensing-Signalwegs (mTOR-Typ)

„Entscheidet", ob eine Zelle wächst und sich teilt oder Ressourcen spart.

Kausaler Beleg (der stärkste auf dieser Liste): Rapamycin, das diesen Signalweg hemmt, verlängerte die Lebensspanne genetisch heterogener Mäuse selbst bei spätem Behandlungsbeginn — ein Meilenstein-Ergebnis aus dem Interventions Testing Program des NIA (Harrison et al., 2009). Ein Regulator der antioxidativen Antwort (Nrf2-Typ)

Steuert das zelleigene Abwehrsystem gegen oxidativen Stress.

Assoziativer Beleg: Die Aktivität dieses Signalwegs nimmt mit dem Alter ab und schwächt die zelluläre Abwehr gegen sich anhäufende oxidative Schäden (Zhang, Davies & Forman, 2015). Indirekter kausaler Beleg: Nacktmulle — Nagetiere mit einer für ihre Größe ungewöhnlich langen Lebensspanne — zeigen eine deutlich höhere Aktivität dieses Signalwegs als gewöhnliche Mäuse (Lewis et al., 2015).

Eine Acetyltransferase, die mit der Autophagie-Regulation verbunden ist

Ein Punkt, an dem die „Aufräumtruppe" der Zelle pharmakologisch angestoßen werden kann.

Assoziativer Beleg: Die autophagische Aktivität im Gewebe nimmt mit dem Alter bei praktisch jedem untersuchten Modellorganismus ab (Rubinsztein, Mariño & Kroemer, 2011). Kausaler Beleg: Mäuse mit künstlich gesteigerter Autophagie (Atg5-Überexpression) leben länger und zeigen bessere Ergebnisse bei der Gewebealterung (Pyo et al., 2013).

Ein Energiesensor (AMPK-Typ)

Wird aktiv, wenn der Zelle Energie fehlt, und löst eine „Energiesparmodus"-Kaskade aus.

Assoziativer Beleg: Die Aktivität dieses Sensors nimmt mit dem Alter in einer Vielzahl von Geweben durchgängig ab (Salminen & Kaarniranta, 2012). Kausaler Beleg: Die durch Kalorienrestriktion bei Fadenwürmern erzeugte Lebensspannenverlängerung erfordert genau diesen Sensor — ohne ihn verschwindet der Effekt (Mair et al., 2011).

Warum das immer noch kein Nachweis für den Nutzen unseres Kandidaten ist

Diese Studien beweisen, dass die Mechanismen selbst valide Ziele der Geroscience sind: Ihre Veränderung kann das Altern von Modellorganismen tatsächlich beeinflussen. Das ist nicht dasselbe wie der Nachweis, dass ein bestimmter Rasayana-Kandidat tatsächlich stark genug in einer lebenden Zelle auf sie wirkt — das ist eine Frage des Dockings (siehe vorherigen Beitrag) und nachfolgender Labortests, nicht allein einer Literaturrecherche.

Warum keine Gennamen. Wie im vorherigen Beitrag sind die Rollen bewusst allgemein beschrieben: Mit dem vollständigen Datensatz „Substanz + Protein + PDB-Struktur" ließe sich unsere Arbeit ohne uns reproduzieren. Die hier zitierten Studien betreffen die Rolle jedes Mechanismus in der Alterungsbiologie im Allgemeinen — unabhängig von Rasayana veröffentlicht und ohne Hinweis darauf, welche konkreten Kandidaten wir testen.
