Ще двадцять років тому «боротьба зі старінням» звучала як маркетингове гасло, а не науковий напрям. Проблема була не в амбіціях, а в означенні: старіння вважали одним суцільним, неподільним процесом — «організм просто зношується». З таким об'єктом важко працювати вченому: незрозуміло, за яку саме ниточку тягнути, що вимірювати, що вважати успіхом втручання.
Перелом стався у 2013 році, коли іспанський біохімік Карлос Лопес-Отін і його колеги — Марія Бласко, Лінда Партрідж, Мануель Серрано, Гвідо Кремер — опублікували в журналі Cell роботу під назвою «The Hallmarks of Aging». Вони розклали старіння на дев'ять окремих, вимірюваних молекулярних процесів — так званих «ознак старіння». У 2023 році та сама команда оновила класифікацію в статті «Hallmarks of aging: An expanding universe», розширивши список до дванадцяти пунктів і розбивши їх на три рівні. Це не філософське спостереження, а практичний інструмент: кожна ознака — конкретний біологічний механізм, який можна виміряти в клітині й, теоретично, на який можна цілеспрямовано впливати.

Первинні ознаки (причина пошкодження)
- Нестабільність геному — накопичення пошкоджень ДНК з роками.
- Вкорочення теломер — захисні «ковпачки» на кінцях хромосом коротшають із кожним поділом клітини.
- Епігенетичні зміни — змінюється не сам текст ДНК, а те, які гени увімкнені чи вимкнені.
- Втрата протеостазу — клітина гірше підтримує правильну форму й якість власних білків.
- Ослаблення макроаутофагії — механізм «прибирання сміття» всередині клітини сповільнюється (див. окрему статтю блогу).
Антагоністичні ознаки (реакція на пошкодження, яка з часом стає шкідливою)
- Дерегуляція чутливості до поживних речовин — шляхи на кшталт AMPK/mTOR, що відчувають рівень енергії в клітині, з віком розбалансовуються.
- Мітохондріальна дисфункція — клітинні «електростанції» працюють гірше, виробляючи більше побічних окислювачів.
- Клітинне старіння (сенесценція) — пошкоджені клітини не гинуть і не діляться, а залишаються й виділяють прозапальні сигнали.
Інтегративні ознаки (наслідки, які зачіпають організм у цілому)
- Виснаження стовбурових клітин — тканини гірше оновлюються й відновлюються.
- Зміна міжклітинної комунікації — клітини гірше «спілкуються» гормональними й хімічними сигналами.
- Хронічне запалення низького ступеня (inflammaging) — фонове запалення, яке не пов'язане з інфекцією.
- Дисбіоз — зміни складу мікробіому кишківника.
Geroscience — це саме та галузь, яка займається не окремими хворобами старості (діабет, серцево-судинні захворювання, деякі види раку) поштучно, а спільним знаменником для них усіх — власне процесом старіння на молекулярному рівні. Логіка проста: якщо вдасться сповільнити один із фундаментальних механізмів, це потенційно відсуне одразу кілька вікових захворювань замість лікування кожного окремо. Саме на цій логіці будуються й реальні клінічні ініціативи — наприклад, запропоноване дослідження TAME (Targeting Aging with Metformin) під керівництвом геронтолога Ніра Барзілая мало б перевірити, чи сповільнює вже давно відомий протидіабетичний препарат метформін вікові процеси в людей без діабету. Інший приклад — рапаміцин, інгібітор шляху mTOR, що послідовно продовжує життя мишам у програмі Interventions Testing Program Національного інституту старіння США; для людини це поки предмет дослідження, а не рекомендація.
Чому це відкриває шлях для обчислювального пошуку речовин
Коли механізм описано на молекулярному рівні — відомо, які саме білки в ньому задіяні — з'являється можливість шукати речовини, що взаємодіють саме з цими білками, ще до дорогих і повільних лабораторних експериментів. Метод називається молекулярний докінг: комп'ютерна симуляція оцінює, наскільки щільно молекула «сідає» в активний центр білка-мішені, за шкалою енергії зв'язування (кілокалорії на моль). Це не заміняє лабораторну перевірку, але дозволяє з тисяч кандидатів відсіяти найперспективніші ще на етапі комп'ютера — саме так побудований дослідницький пайплайн, який лежить в основі проєкту Rasayana (детальніше — на сторінці «Лабораторія»).
Важливо чесно проговорити межі методу: сильний результат докінгу — це гіпотеза, а не доказ ефективності. Наступні кроки — перевірка на клітинних культурах (in vitro), потім на тваринних моделях, і лише згодом, за багато років, — клінічні дослідження на людях. Жодна речовина в цьому процесі не стає «ліками від старості» одразу після симуляції на комп'ютері.

Twenty years ago, "fighting aging" sounded like a marketing slogan rather than a scientific field. The problem wasn't ambition — it was definition: aging was treated as one indivisible process, as if "the body just wears out." That's a hard thing for a scientist to work with — there's no clear thread to pull, nothing specific to measure, no clear definition of what a successful intervention would even look like.
The turning point came in 2013, when Spanish biochemist Carlos López-Otín and his colleagues — María Blasco, Linda Partridge, Manuel Serrano, Guido Kroemer — published a paper in Cell titled "The Hallmarks of Aging." They broke aging down into nine distinct, measurable molecular processes — the so-called hallmarks of aging. In 2023 the same team updated the framework in "Hallmarks of aging: An expanding universe," expanding the list to twelve and organizing them into three tiers. This isn't a philosophical observation — it's a practical tool: each hallmark is a concrete biological mechanism that can be measured in a cell and, in principle, targeted deliberately.
The twelve hallmarks of aging — the 2023 structure
Primary hallmarks (the root causes of damage)
- Genomic instability — DNA damage accumulates over the years.
- Telomere attrition — the protective "caps" on the ends of chromosomes shorten with every cell division.
- Epigenetic alterations — not the DNA text itself changing, but which genes are switched on or off.
- Loss of proteostasis — the cell gets worse at maintaining the correct shape and quality of its own proteins.
- Disabled macroautophagy — the cell's internal "cleanup" mechanism slows down (see the separate blog post on this).
Antagonistic hallmarks (a response to damage that turns harmful over time)
- Deregulated nutrient sensing — pathways like AMPK/mTOR, which sense the cell's energy state, become imbalanced with age.
- Mitochondrial dysfunction — the cell's "power plants" work less efficiently, producing more byproduct oxidants.
- Cellular senescence — damaged cells stop dividing but don't die either, lingering and releasing pro-inflammatory signals.
Integrative hallmarks (consequences affecting the organism as a whole)
- Stem cell exhaustion — tissues become worse at renewing and repairing themselves.
- Altered intercellular communication — cells get worse at "talking" to each other via hormonal and chemical signals.
- Chronic low-grade inflammation (inflammaging) — background inflammation unrelated to any infection.
- Dysbiosis — shifts in the composition of the gut microbiome.
Geroscience is precisely the field that doesn't chase individual diseases of old age (diabetes, cardiovascular disease, certain cancers) one at a time, but their common denominator — the aging process itself, at the molecular level. The logic is simple: slow down one of the fundamental mechanisms, and you potentially push back several age-related diseases at once, instead of treating them one by one. Real clinical initiatives are built on exactly this logic — the proposed TAME trial (Targeting Aging with Metformin), led by gerontologist Nir Barzilai, aims to test whether a decades-old diabetes drug, metformin, slows aging processes in people who don't have diabetes. Another example is rapamycin, an mTOR-pathway inhibitor that has consistently extended lifespan in mice through the US National Institute on Aging's Interventions Testing Program; for humans, this remains a research question, not a recommendation.
Why this opens the door to computational compound discovery
Once a mechanism is described at the molecular level — once you know which specific proteins are involved — it becomes possible to search for compounds that interact with those exact proteins, before any expensive, slow lab work begins. The method is called molecular docking: a computer simulation estimates how tightly a molecule "fits" into a target protein's active site, on a binding-energy scale measured in kilocalories per mole. It doesn't replace lab validation, but it lets you narrow thousands of candidates down to the most promising ones at the computer stage — which is exactly how the research pipeline behind the Rasayana project works (more on the Lab page).
It's worth being honest about the method's limits: a strong docking result is a hypothesis, not proof of effectiveness. The next steps are testing in cell cultures (in vitro), then in animal models, and only much later — after many years — human clinical trials. No compound becomes an "anti-aging drug" the moment a computer simulation looks promising.

Vor zwanzig Jahren klang „Alterskampf" eher nach Marketing-Slogan als nach Wissenschaftszweig. Das Problem war nicht der Ehrgeiz, sondern die Definition: Altern galt als ein einziger, unteilbarer Prozess — als würde „der Körper einfach verschleißen". Damit lässt sich schwer arbeiten: Es ist unklar, an welchem Faden man ziehen soll.
Die Wende kam 2013, als eine Forschungsgruppe (López-Otín und Kollegen) eine Arbeit veröffentlichte, die das Altern in neun eigenständige, messbare molekulare Prozesse zerlegte — die sogenannten „Hallmarks of Aging". 2023 wurde die Liste aktualisiert und auf zwölf erweitert. Das ist keine philosophische Beobachtung, sondern ein praktisches Werkzeug: Jedes Kennzeichen ist ein konkreter biologischer Mechanismus, der in einer Zelle gemessen und im Prinzip mit einem Wirkstoff angesteuert werden kann.
Ein paar Beispiele für Alterungskennzeichen
- Telomerverkürzung — die schützenden „Kappen" an den Enden der Chromosomen werden mit jeder Zellteilung kürzer.
- Mitochondriale Dysfunktion — die „Kraftwerke" der Zelle arbeiten mit dem Alter weniger effizient und produzieren mehr oxidative Nebenprodukte.
- Nachlassende Autophagie — der interne „Aufräum"-Mechanismus der Zelle verlangsamt sich (mehr dazu in einem separaten Blogbeitrag).
- Zelluläre Seneszenz — beschädigte Zellen teilen sich nicht mehr, sterben aber auch nicht, verbleiben und senden entzündungsfördernde Signale aus.
- Deregulierte Nährstofferkennung — Signalwege wie AMPK/mTOR, die den Energiezustand der Zelle erfassen, geraten mit dem Alter aus dem Gleichgewicht.
Geroscience ist genau das Feld, das nicht einzelne Alterskrankheiten (Diabetes, Herz-Kreislauf-Erkrankungen, bestimmte Krebsarten) einzeln angeht, sondern deren gemeinsamen Nenner — den Alterungsprozess selbst, auf molekularer Ebene. Die Logik ist einfach: Verlangsamt man einen der grundlegenden Mechanismen, verschiebt man potenziell mehrere altersbedingte Erkrankungen gleichzeitig, statt sie einzeln zu behandeln.
Warum das den Weg für computergestützte Wirkstoffsuche öffnet
Sobald ein Mechanismus auf molekularer Ebene beschrieben ist — sobald bekannt ist, welche konkreten Proteine beteiligt sind — wird es möglich, nach Substanzen zu suchen, die genau mit diesen Proteinen interagieren, noch bevor teure, langsame Laborarbeit beginnt. Die Methode heißt molekulares Docking: Eine Computersimulation schätzt, wie gut ein Molekül in die aktive Tasche eines Zielproteins „passt". Sie ersetzt keine Laborvalidierung, erlaubt aber, aus Tausenden Kandidaten die vielversprechendsten bereits am Computer herauszufiltern — genau so ist die Forschungs-Pipeline hinter dem Rasayana-Projekt aufgebaut (mehr auf der Labor-Seite).
Man sollte ehrlich über die Grenzen der Methode sprechen: Ein starkes Docking-Ergebnis ist eine Hypothese, kein Wirksamkeitsnachweis. Die nächsten Schritte sind Tests in Zellkulturen (in vitro), dann in Tiermodellen, und erst viel später — nach vielen Jahren — klinische Studien am Menschen. Keine Substanz wird allein durch eine vielversprechende Computersimulation zu einem „Anti-Aging-Medikament".
