У попередній статті блогу ми пояснювали, що geroscience розклала старіння на конкретні, вимірювані молекулярні механізми — «ознаки старіння». Наступний логічний крок: якщо механізм описано на рівні конкретних білків, можна шукати речовини, що на ці білки впливають, ще до дорогих лабораторних експериментів. Це і є молекулярний докінг — метод, на якому побудована обчислювальна частина Rasayana.

У нашому пайплайні три кандидати (відібрані з 39 традиційних рецептів) перевірено проти шести білків-мішеней старіння — 18 пар «кандидат×мішень» усього, найкраща передбачена афінність зв'язування склала −9,7 ккал/моль. Ці цифри вже публічні (див. головну сторінку). А от самі назви речовин і конкретні білки — ні: маючи повний набір даних, нашу роботу можна було б відтворити без нас, тож ми свідомо публікуємо тут лише ролі мішеней, без назв генів чи PDB-кодів.

Шість ролей, які ми перевіряємо

Фермент підтримки теломер

Добудовує захисні «ковпачки» на кінцях хромосом, які коротшають з кожним поділом клітини — один із класичних маркерів клітинного старіння.

Що вважали б доказом користі: реальне уповільнення вкорочення теломер у клітинній культурі (вимірюється qPCR або Flow-FISH), а не лише сильний докінг-скор.

NAD⁺-залежна деацетилаза (сиртуїновий тип)

Регулює метаболічну «економність» клітини у відповідь на рівень енергії; активність цього класу білків знижується з віком паралельно з падінням рівня NAD⁺.

Що вважали б доказом користі: зростання співвідношення NAD⁺/NADH або пряме підвищення ферментативної активності в лізаті клітин після впливу речовини.

Кіназа шляху чутливості до поживних речовин (mTOR-типу)

Цей шлях «вирішує», чи клітина росте й ділиться, чи економить ресурси; хронічна гіперактивність пов'язана з пришвидшеним старінням, а часткове пригнічення — з подовженням життя у тваринних моделях.

Що вважали б доказом користі: зниження фосфорилювання низхідних мішеней цього шляху (вимірюється Western blot) без токсичності для клітини.

Регулятор антиоксидантної відповіді (Nrf2-типу)

Керує «увімкненням» власної системи захисту клітини від окислювального стресу — одного з накопичувальних факторів пошкодження з віком.

Що вважали б доказом користі: активація ARE-репортерної системи або зниження маркерів окислювального пошкодження (наприклад, 8-OHdG) в обробленій культурі.

Ацетилтрансфераза, пов'язана з регуляцією автофагії

Автофагія — «служба прибирання сміття» клітини (детальніше — в окремій статті блогу); цей фермент — одна з точок, де його можна фармакологічно підкрутити.

Що вважали б доказом користі: зміна співвідношення LC3-II/p62 — стандартного маркера автофагічного потоку — у бік активнішого «прибирання».

Енергетичний сенсор (AMPK-типу)

Активується, коли клітині бракує енергії, і запускає каскад «режиму економії» — шлях, з яким пов'язують ефекти фізичних вправ і обмеження калорій на довголіття.

Що вважали б доказом користі: зростання співвідношення AMP/ATP разом із фосфорилюванням низхідних мішеней сенсора.

Докінг — це гіпотеза, а не результат

Важливо чесно розмежувати два різні твердження. Перше: «речовина X передбачено добре сідає в кишеню білка Y» — це те, що дає нам молекулярний докінг, і саме це ми вже маємо для всіх 18 пар. Друге: «речовина X реально уповільнює процес, за який відповідає білок Y» — це можна показати лише експериментально, на клітинній культурі, а потім на тваринних моделях. Ми поки на етапі першого твердження й відкрито про це говоримо: сильний докінг-скор — це підстава перевіряти далі, а не готовий результат.

Саме тому наступний крок проєкту — не маркетинг, а домовленість із лабораторією про перелічені вище тести in vitro: цитотоксичність, а потім — конкретні функціональні аналізи під кожну мішень і кожен біомаркер зі списку вище. Коли з'являться перші реальні (а не передбачені) цифри, ми розповімо про це в блозі так само відкрито.

Чому без назв. Ролі мішеней описано узагальнено навмисно, так само як на головній сторінці ми не називаємо самі речовини: маючи повний набір «речовина + білок + PDB-структура», нашу роботу можна було б відтворити без нас. Повне досьє розкривається серйозним інвесторам після розмови, під NDA.

In an earlier post we explained how geroscience broke aging down into concrete, measurable molecular mechanisms — the "hallmarks of aging." The next logical step: once a mechanism is described at the level of specific proteins, you can search for compounds that act on those proteins before any expensive lab work begins. That's molecular docking — the method behind Rasayana's computational pipeline.

In our pipeline, three candidates (shortlisted from 39 traditional recipes) were tested against six aging-related protein targets — 18 candidate×target pairs in total, with the best predicted binding affinity at −9.7 kcal/mol. Those numbers are already public (see the homepage). The compound names and the specific proteins are not: with the full dataset, our work could be reproduced without us, so here we deliberately publish only the targets' general roles, without gene names or PDB codes.

The six roles we test against

A telomere-maintenance enzyme

Rebuilds the protective "caps" on chromosome ends, which shorten with every cell division — one of the classic markers of cellular aging.

What would count as evidence of benefit: a measurable slowdown in telomere shortening in cell culture (via qPCR or Flow-FISH), not just a strong docking score.

An NAD⁺-dependent deacetylase (sirtuin-type)

Regulates the cell's metabolic "thriftiness" in response to energy status; this class of protein's activity declines with age alongside falling NAD⁺ levels.

What would count as evidence of benefit: a rise in the NAD⁺/NADH ratio, or a direct increase in enzymatic activity in cell lysate after exposure to the compound.

A nutrient-sensing pathway kinase (mTOR-type)

This pathway "decides" whether a cell grows and divides or conserves resources; chronic hyperactivity is linked to accelerated aging, while partial inhibition extends lifespan in animal models.

What would count as evidence of benefit: reduced phosphorylation of downstream targets in this pathway (measured by Western blot), without cellular toxicity.

An antioxidant-response regulator (Nrf2-type)

Controls the cell's own defense system against oxidative stress — one of the cumulative damage factors that builds up with age.

What would count as evidence of benefit: activation of an ARE-reporter system, or reduced oxidative-damage markers (e.g. 8-OHdG) in treated culture.

An acetyltransferase linked to autophagy regulation

Autophagy is the cell's "cleanup crew" (see our separate post on it); this enzyme is one point where it can be pharmacologically nudged.

What would count as evidence of benefit: a shift in the LC3-II/p62 ratio — the standard marker of autophagic flux — toward more active cleanup.

An energy sensor (AMPK-type)

Activates when a cell is short on energy and triggers an "energy-saving mode" cascade — a pathway linked to the longevity effects of exercise and caloric restriction.

What would count as evidence of benefit: a rise in the AMP/ATP ratio together with phosphorylation of the sensor's downstream targets.

Docking is a hypothesis, not a result

It's worth being precise about two very different claims. The first: "compound X is predicted to fit well into protein Y's pocket" — that's what molecular docking gives us, and we already have that for all 18 pairs. The second: "compound X actually slows the process protein Y is responsible for" — that can only be shown experimentally, first in cell culture and then in animal models. We're currently at the first claim, and we say so openly: a strong docking score is a reason to test further, not a finished result.

That's exactly why the project's next step isn't marketing — it's arranging the in vitro tests listed above with a lab: cytotoxicity screening first, then specific functional assays for each target and each biomarker above. When the first real (not predicted) numbers come in, we'll report them here just as openly.

Why no names. Target roles are described generically on purpose, the same way we don't name the compounds themselves on the homepage: with the full "compound + protein + PDB structure" dataset, our work could be reproduced without us. The full dossier is shared with serious investors after a conversation, under NDA.

Das ist molekulares Docking, die Methode hinter der computergestützten Pipeline von Rasayana.

In unserer Pipeline wurden drei Kandidaten (aus 39 traditionellen Rezepturen vorausgewählt) gegen sechs alterungsrelevante Zielproteine getestet — insgesamt 18 Kandidat×Zielprotein-Paare, mit einer besten vorhergesagten Bindungsaffinität von −9,7 kcal/mol. Diese Zahlen sind bereits öffentlich (siehe Startseite). Die Substanznamen und die konkreten Proteine sind es nicht: Mit dem vollständigen Datensatz ließe sich unsere Arbeit ohne uns reproduzieren, deshalb veröffentlichen wir hier bewusst nur die allgemeinen Rollen der Zielproteine, ohne Gennamen oder PDB-Codes.

Die sechs Rollen, gegen die wir testen

Ein Enzym zur Telomer-Erhaltung

Baut die schützenden „Kappen" an den Chromosomenenden wieder auf, die sich mit jeder Zellteilung verkürzen — eines der klassischen Kennzeichen zellulärer Alterung.

Was als Wirksamkeitsnachweis zählen würde: eine messbare Verlangsamung der Telomerverkürzung in Zellkultur (per qPCR oder Flow-FISH), nicht nur ein starker Docking-Score.

Eine NAD⁺-abhängige Deacetylase (Sirtuin-Typ)

Reguliert die metabolische „Sparsamkeit" der Zelle je nach Energiestatus; die Aktivität dieser Proteinklasse sinkt mit dem Alter parallel zum fallenden NAD⁺-Spiegel.

Was als Wirksamkeitsnachweis zählen würde: ein Anstieg des NAD⁺/NADH-Verhältnisses oder eine direkt erhöhte Enzymaktivität im Zelllysat nach Substanzeinwirkung.

Eine Kinase des Nährstoff-Sensing-Signalwegs (mTOR-Typ)

Dieser Signalweg „entscheidet", ob eine Zelle wächst und sich teilt oder Ressourcen spart; chronische Überaktivität ist mit beschleunigter Alterung verknüpft, eine teilweise Hemmung verlängert in Tiermodellen die Lebensspanne.

Was als Wirksamkeitsnachweis zählen würde: verringerte Phosphorylierung nachgeschalteter Zielproteine dieses Signalwegs (per Western Blot), ohne Zelltoxizität.

Ein Regulator der antioxidativen Antwort (Nrf2-Typ)

Steuert die „Aktivierung" des zelleigenen Abwehrsystems gegen oxidativen Stress — einen der Schadensfaktoren, die sich mit dem Alter kumulativ aufbauen.

Was als Wirksamkeitsnachweis zählen würde: Aktivierung eines ARE-Reportersystems oder reduzierte oxidative Schadensmarker (z. B. 8-OHdG) in behandelter Kultur.

Eine Acetyltransferase, die mit der Autophagie-Regulation verbunden ist

Autophagie ist die „Aufräumtruppe" der Zelle (siehe unseren separaten Beitrag dazu); dieses Enzym ist einer der Punkte, an denen sie pharmakologisch angestoßen werden kann.

Was als Wirksamkeitsnachweis zählen würde: eine Verschiebung des LC3-II/p62-Verhältnisses — dem Standardmarker für den autophagischen Fluss — hin zu aktiverem Aufräumen.

Ein Energiesensor (AMPK-Typ)

Wird aktiv, wenn der Zelle Energie fehlt, und löst eine „Energiesparmodus"-Kaskade aus — ein Signalweg, der mit den Langlebigkeitseffekten von Bewegung und Kalorienrestriktion in Verbindung gebracht wird.

Was als Wirksamkeitsnachweis zählen würde: ein Anstieg des AMP/ATP-Verhältnisses zusammen mit der Phosphorylierung nachgeschalteter Zielproteine des Sensors.

Docking ist eine Hypothese, kein Ergebnis

Zwei sehr unterschiedliche Aussagen sollte man klar trennen. Die erste: „Substanz X passt vorhergesagt gut in die Tasche von Protein Y" — das liefert molekulares Docking, und das haben wir bereits für alle 18 Paare. Die zweite: „Substanz X verlangsamt tatsächlich den Prozess, für den Protein Y verantwortlich ist" — das lässt sich nur experimentell zeigen, zunächst in Zellkultur, dann in Tiermodellen. Wir befinden uns aktuell bei der ersten Aussage, und das sagen wir offen: Ein starker Docking-Score ist ein Grund für weitere Tests, kein fertiges Ergebnis.

Genau deshalb ist der nächste Schritt des Projekts kein Marketing, sondern die Abstimmung der oben genannten In-vitro-Tests mit einem Labor: zunächst Zytotoxizitäts-Screening, danach spezifische funktionelle Assays für jedes Zielprotein und jeden Biomarker oben. Sobald erste echte (nicht vorhergesagte) Zahlen vorliegen, berichten wir hier genauso offen darüber.

Warum keine Namen. Die Rollen der Zielproteine sind absichtlich allgemein beschrieben, genau wie wir auf der Startseite die Substanzen selbst nicht benennen: Mit dem vollständigen Datensatz „Substanz + Protein + PDB-Struktur" ließe sich unsere Arbeit ohne uns reproduzieren. Das vollständige Dossier wird ernsthaften Investoren nach einem Gespräch unter NDA zugänglich gemacht.
